Introduction: Guanidine isothiocyanate is in white crystalline form, with melting point of 116~121 ℃. At 20 ℃, the PH value of 6mol/L in water is 4.5~6, and the solubility is complete dissolution. It is a kind of decoupling agent, a kind of strong protein denaturant, which can dissolve the protein, make the secondary structure of the protein disappear, degrade the cell structure, and rapidly separate the nuclear protein from the nucleic acid. Used for denatured lysed cells; Rna and dna were extracted without rnase and dnase activity

Use: Guanidine thiocyanate can be used as biochemical reagent and pharmaceutical intermediate. It is used for degreasing in oil and fat industry, as a strong fat degradation agent in daily chemical industry, and as a cell membrane breaking agent in biopharmaceuticals.

Principle of action: the principle of guanidine isothiocyanate-phenol method to extract rna: guanidine isothiocyanate and b-mercaptoethanol work together to inhibit the activity of rnase, while guanidine isothiocyanate and sodium dodecyl sarcosine work together to denature the protein and release rna; Under acidic conditions, dna rarely dissociates, denatures with protein and precipitates, while rna dissolves in supernatant.

Research purpose: guanidine thiocyanate has been used: • Splitting colorectal adenocarcinoma cell lines before RNA separation • As a PCR inhibitor for triple mitochondrial DNA (mtDNA) qPCR detection, guanidine thiocyanate is a denaturant, commonly used for RNA separation. It is used as a storage buffer for whole blood samples. Guanidine thiocyanate can inactivate nuclease and is an ideal choice for storing and freezing fecal samples for DNA research. Used in combination with phenol-chloroform in RNA extraction
